PBPK (Physiologically Based Pharmacokinetic Modeling)

At a glance: A mechanistic model predicting how chemicals move through the body over time.

PBPK modeling is a mathematical technique for predicting the absorption, distribution, metabolism, and excretion of chemical substances in humans and other species. It is used in drug development and in health risk assessment for cosmetics and general chemicals. 61 Unlike simpler compartmental models, a PBPK model simulates how a drug’s concentration changes over time in tissues and blood through the interaction of physiological, physicochemical, and biochemical determinants. 62 PBPK supplies internal-dose estimates that connect external exposure to bioactivity in NGRA and in-vitro-to-in-vivo extrapolation. It is closely related to PBK modelling.
